No, this does not happen. As Al says, even if water and ethanol weren’t highly miscible, ethanol is much less dense than water and would tend to float on top of water, not sink to the bottom. Even a denser sugar heavy liqueur is going to mix well in a drink. The only thing that will settle out are larger, non- molecular solids, like the remains of any blended fruits.
